10. Discovery and Characterization of Novel Interactions Involving PrP and Related Proteins

Project Description: This project aims to increase our understanding of the function of the prion protein (PrPC) and the biogenesis of its infectious form (PrPSc) by clarifying the other proteins that PrPC contacts in the cell membrane. Building on previous research that employed the novel use of yeast prions to identify proteins expressed in the human brain that interact with human PrP, this technology will be used to discover proteins that interact with PrP-related proteins Shadoo and Doppel. Tissue culture cells will also be used to investigate selected interactions in more detail and to probe the functional implications of these interactions in normal and diseased states.
